Research summary: Modulation of immune responses to alleviate diseases has been of interest for many years and concept of rasayana in Ayurveda is based on same principles. Various botanicals mentioned under rasayana category of therapeutics are prescribed for rejuvenation. Further, formulations prepared from Rasayana plants are reported to have immunomodulatory potential. This

stimulated us to explore their potential in adjuvant discovery. Tinospora cordifolia [TC] - a Rasayana botanical was shortlisted for the study.

Optimization for sensitization/ immunization and challenge dose. Measurement of immune cell populations (CD3+, CD19+ & CD4+, CD8+]. Signature cytokines [IL-2, IFN-gamma, IL-4 and IL-10] counting. Antigen specific immune responses. B. Develop and standardize in-vivo guinea pig model to bacterial (DPT) antigens for protective

efficacy of immunomodulators [host challenge method]. Optimization of effective vaccine dilution. Effective treatment regimen. Challenge dose optimization with respect to protective efficacy. Mortality and Morbidity observations i.e. challenge associated death, devised scoring scheme for

symptoms, adrenal hemorrhages and sera cortisol levels. C. Develop and standardize in-vivo animal models to polysaccharide based (Men. A) antigens for

screening of immunomodulators in mice. Optimization of effective treatment regimen. Booster dose minimization. D. Other animal experimental techniques: expertise with intravenous, intramuscular, intraperitoneal

and oral dosing to the laboratory animals and experience in small animal surgeries such as isolation of Adrenals, spleen, Kidney, liver and heart.

E. Molecular Biology: Demonstrated experience with cellular and molecular immunological assays such as assessment of

humoral and cellular immunity markers using ELISA, flow cytometry [lymphocyte phenotyping and intracellular cytokine estimations] and proliferation assays.
